Fluorescence emission is enhanced upon insertion of the reactive center loop into beta-sheet A (following the formation of complexes with proteinases, formation of the latent species, or cleavage by elastase). This modified version of PAI-1 is nearly as active as wild type PAI-1, but is more resistant to spontaneous latency reaction (making it suitable in applications like monitoring reaction rates of PAI-1).
